About the Trial

OlympiA randomized 1,836 patients with germline BRCA1 or BRCA2 mutations and high risk HER2 negative early breast cancer to adjuvant olaparib for a year or placebo. The PARP inhibitor reduced invasive disease events by 42 percent and death by 32 percent. The trial moved PARP inhibition from metastatic to curative settings and entrenched germline genetic testing in early breast cancer management. It also validated the synthetic lethality concept, tumor BRCA deficiency plus PARP blockade, in the curative setting. OlympiA exemplifies how germline genomics now personalizes adjuvant therapy decisions.
