About the Trial

The Multicenter International Study of Oxaliplatin 5 Fluorouracil Leucovorin in the Adjuvant Treatment of Colon Cancer randomized 2,246 stage II and III colon cancer patients to FOLFOX or standard 5 FU leucovorin. FOLFOX improved 3 year disease free survival from 72.9 to 78.2 percent, later translating into overall survival gain in stage III. Oxaliplatin, a platinum analog with distinctive neurotoxicity, became the third drug in colorectal therapy. Cure rates for stage III colon cancer rose by roughly 5 absolute percent. MOSAIC set the adjuvant backbone that the IDEA collaboration later refined by studying treatment duration, and it remains foundational in colon cancer management.
