About the Trial
The International Randomized Study of Interferon and STI571 randomized 1,106 newly diagnosed chronic phase CML patients to imatinib or interferon plus cytarabine. Imatinib was so superior, about 95 percent complete hematologic response and 76 percent complete cytogenetic response at 18 months, that crossover was massive and the comparison effectively ended. Ten year follow up showed an estimated 83 percent survival, with most patients living normal lives on a pill. Imatinib, developed by Druker and Lydon, was the first targeted therapy designed from a molecular lesion, the BCR ABL kinase. It founded precision oncology and received FDA approval in 2001 after an unusually fast review.
