Key FindingGefitinib beats chemotherapy in EGFR mutated lung cancer

About the Trial

The Iressa Pan Asia Study randomized 1,217 East Asian never or light smokers with lung adenocarcinoma to gefitinib or carboplatin paclitaxel. Gefitinib improved progression free survival overall, but the effect depended on mutation status: among the roughly 60 percent of tested tumors with EGFR mutations, gefitinib roughly halved the risk of progression, while in wild type tumors chemotherapy did better. This prespecified biomarker analysis proved targeted therapy requires the target. IPASS, with NEJ002 and EURTAC, established routine EGFR mutation testing in lung adenocarcinoma and started the molecular stratification of lung cancer that now includes ALK, ROS1, RET, MET, and more. Lung oncology became the paradigm of genomically driven treatment.
