About the Trial

The Herceptin Adjuvant trial randomized 5,102 women with HER2 positive early breast cancer to trastuzumab for 1 or 2 years or observation after chemotherapy. One year of trastuzumab reduced disease free survival events by 46 percent and death by about a third, one of the largest benefits ever seen in adjuvant breast oncology. The 2 year arm added nothing. Cardiac toxicity was real but manageable. HERA, with the concurrent NSABP B-31 and N9831 trials, transformed HER2 positive disease from the worst to one of the most treatable breast cancer subtypes, a triumph of targeted therapy driven by Dennis Slamon's work linking HER2 amplification to aggressive disease.
