About the Trial
FLAURA randomized 556 patients with untreated EGFR mutated advanced non small cell lung cancer to third generation osimertinib or first generation EGFR inhibitors. Osimertinib extended progression free survival to 18.9 versus 10.2 months and later showed overall survival benefit, 38.6 versus 31.8 months, an overall survival advantage over earlier generation EGFR TKIs. It penetrates the brain, cutting CNS progression, and spares wild type EGFR, reducing rash and diarrhea. Osimertinib became a widely adopted first line treatment. The trial completed a 20 year arc from gefitinib discovery through resistance mutation T790M exploitation, one of oncology most elegant targeted therapy sequences.
