About the Trial

The DPP randomized 3,234 people with impaired glucose tolerance to metformin, intensive lifestyle modification, or placebo. Lifestyle intervention, targeting 7 percent weight loss and 150 minutes weekly exercise, reduced diabetes incidence by 58 percent. Metformin reduced it by 31 percent. Lifestyle worked in every subgroup and was far more effective than metformin in adults aged 60 and over. The trial was stopped about a year early once the benefit was clear. In DPPOS follow up, participants who did not develop diabetes had fewer microvascular complications. The DPP lifestyle program has been translated into the CDC National Diabetes Prevention Program used across the United States today.
